Every Cambridge IGCSE Biology (0610) candidate sits three papers: a multiple-choice paper, a theory paper and a practical paper. Core candidates take Papers 1 and 3, Extended candidates take Papers 2 and 4, and both tiers add either Paper 5 (Practical Test) or Paper 6 (Alternative to Practical). Knowing how the Cambridge IGCSE Biology 0610 papers fit together, from timings and marks to the skills each one rewards, turns a long topic list into a revision plan you can actually follow. (This guide has been updated for the 2026-27 academic year.)

Key Takeaways

Core candidates sit Papers 1 and 3 and are eligible for grades C to G, while Extended candidates sit Papers 2 and 4 and are eligible for grades A* to G.
Theory is worth the most: Paper 3 or Paper 4 is 80 marks and 50% of the qualification, against 30% for the multiple-choice paper and 20% for the practical paper.
Every candidate also takes one practical paper, either Paper 5 (Practical Test, in a laboratory) or Paper 6 (Alternative to Practical, with no experiments to carry out), and both test experimental skills only.
Calculators may be used in all parts of the exam, so the maths you need (percentages, means, standard form, magnification) is a skill to practise, not a reason to worry.
Candidates taught only the Core content can reach a maximum of grade C, so anyone aiming for A* to C should study the Extended content.

The Cambridge IGCSE Biology 0610 Papers at a Glance

The current 0610 syllabus is for exams in 2026, 2027 and 2028, so it is the one that applies to students sitting the exam during the 2026-27 academic year. Cambridge has also published a separate syllabus for 2029 exams, and its assessment structure is the same. Exams run in the June and November series, with the March series available in India. All the papers are externally assessed and every question is compulsory, so there is no choice of questions and no coursework.

Paper	Who takes it	Format	Duration	Marks	Weighting
Paper 1	Core	Multiple choice: 40 four-option questions	45 minutes	40	30%
Paper 2	Extended	Multiple choice: 40 four-option questions	45 minutes	40	30%
Paper 3	Core	Theory: short-answer and structured questions	1 hour 15 minutes	80	50%
Paper 4	Extended	Theory: short-answer and structured questions	1 hour 15 minutes	80	50%
Paper 5	Core and Extended (one practical paper)	Practical Test, done in a laboratory	1 hour 15 minutes	40	20%
Paper 6	Core and Extended (one practical paper)	Alternative to Practical, no experiments to carry out	1 hour	40	20%

In plain terms, your three papers are a multiple-choice paper and a theory paper for your tier, plus one of the two practical papers. The odd-numbered papers are Core and the even-numbered papers are Extended, while Papers 5 and 6 are shared by both tiers. The questions in Papers 1 and 3 are based on the Core content only, and the questions in Papers 2 and 4 are based on the Core and Supplement content together.

If the jump from topic notes to three different paper styles feels like a lot, a tutor can build a plan around your tier and your weakest topics from the first session. Tell us what you need help with →

Core vs Extended: Which Tier Fits You?

The tier sets the papers you sit and the grades you can reach. According to the syllabus, candidates who have studied the Core content, or who are expected to achieve a grade D or below, should be entered for Papers 1 and 3, and they are eligible for grades C to G. Candidates who have studied the Extended content (the Core plus the Supplement) and who are expected to achieve a grade C or above should be entered for Papers 2 and 4, and they are eligible for grades A* to G. Cambridge adds that candidates taught only the Core content can achieve a maximum of grade C, and that those aiming for grades A* to C should be taught the Extended content.

The Core and Supplement content are laid out side by side across 21 topics, from characteristics and classification of living organisms, through enzymes, plant and human nutrition, respiration and inheritance, to human influences on ecosystems and biotechnology and genetic modification. The Supplement column is where the extra depth sits, so the tier decision is really a decision about how far into each topic you will be examined.

The two multiple-choice papers and the two theory papers also test the same two skills in the same proportions. Within Papers 1 to 4, 63% of the marks go to AO1 (knowledge with understanding) and 37% to AO2 (handling information and problem-solving). Across the whole qualification, the approximate weightings are 50% AO1, 30% AO2 and 20% AO3 (experimental skills and investigations), and the practical papers are where all of the AO3 marks sit. In practice that means more than a third of the marks in your multiple-choice and theory papers reward using what you know on unfamiliar information, such as graphs, tables, diagrams and new scenarios, not just recalling it.

Talk to your teacher early about which tier you are being entered for, because the decision changes the content you need to revise and the grades available to you.

Paper 5 or Paper 6: The Practical Papers

Every candidate takes one practical paper from a choice of two, and both are worth 40 marks and 20% of the qualification. The Practical Test (Paper 5) lasts 1 hour 15 minutes and requires candidates to do experiments in a laboratory. The Alternative to Practical (Paper 6) lasts 1 hour and does not require candidates to carry out experiments. Cambridge states that the two papers require the same experimental skills and the same understanding of experimental contexts, and that both test AO3. Ask your teacher which of the two you are entered for, because the preparation differs a little.

Candidates are expected to be familiar with, and may be asked about, a defined list of experimental contexts:

Simple quantitative experiments measuring volumes, masses, temperatures, times and lengths
Diffusion and osmosis
Food tests
Rates of enzyme-catalysed reactions, including judging end-points such as colour changes
pH and the use of hydrogencarbonate indicator, litmus and universal indicator
Photosynthesis (rate and limiting factors) and transpiration
Heart rate and breathing rate, and respiration
Tropic responses, germination, and the observation and dissection of seeds and flowers
Continuous and discontinuous variation, and sampling methods that avoid bias
Observing and drawing biological specimens, including calculating magnification or actual size

The skills behind these experiments are the same whichever paper you take. You may be asked to plan an investigation, identifying the independent and dependent variables, describing which variables to keep constant, and suggesting a suitable control. You may also be asked to record results in a table, plot a graph, spot anomalous results, draw a conclusion and evaluate the method, including sources of error and possible improvements. Because candidates taking Paper 6 do not carry out experiments, they still need to learn these techniques properly; they show them in answers rather than at the bench.

How to Prepare for Each Paper

Multiple choice (Paper 1 or Paper 2)

You have 45 minutes for 40 four-option questions, which is a little over a minute per question. These papers draw on the subject content for your tier, so breadth across all 21 topics matters. Revise topic by topic, then mix topics together in timed sets so you practise switching between, say, enzymes and ecosystems. When you review wrong answers, write down why each wrong option was tempting, because a pattern in your mistakes is the quickest thing to fix.

Theory (Paper 3 or Paper 4)

Theory is the biggest single block: 80 marks in 1 hour 15 minutes, which works out at slightly less than a minute per mark. The questions are short-answer and structured, so a clear, precise sentence usually earns more than a long paragraph. Learn the command words Cambridge lists for this syllabus, because each asks for something different:

State and give want a clear answer from recall, with no explanation needed.
Describe asks for the main features or points of a topic.
Explain asks for purposes or reasons, saying why and how, supported with relevant evidence.
Compare asks you to comment on similarities and/or differences, so name both sides.
Suggest asks you to apply knowledge to a situation where there is a range of valid responses, as in unfamiliar-context questions.
Calculate and determine ask you to work something out from given information, so show your steps.

Because about 37% of the marks in the written papers go to handling information and problem-solving, practise with data. Our guide to mastering biology data-based questions walks through reading graphs and tables step by step, and the same habits apply to IGCSE questions.

Maths you can use a calculator for

The syllabus states that calculators may be used in all parts of the exam. The mathematical skills it lists include working with percentages and percentage change, standard form, significant figures and decimal places, means and ranges, interpolating and extrapolating from data, and converting between units such as mm, cm and m. It also lists surface area to volume ratio and the area of a rectangle, triangle and circle, so those calculations are fair game. Practise each one with real biology data rather than abstract numbers.

Practical skills (Paper 5 or Paper 6)

Cambridge publishes clear conventions for presenting data, and following them is the easiest way to protect marks:

Head each table column with the quantity and unit separated by a solidus, for example time / s, and keep units out of the body of the table.
Plot the independent variable on the x-axis and the dependent variable on the y-axis unless told otherwise, mark points as crosses or encircled dots, and plot each point to within half of the smallest square.
Draw a single, thin, smooth best-fit line or curve with a sharp pencil, ignoring points that are clearly anomalous.
Draw bar charts with bars that do not touch for categorical or discrete data, and histograms with bars that touch for continuous data.
Draw biological specimens in sharp pencil with clear, unbroken lines, using most of the available space, with no shading or colour, and draw label lines with a ruler so they touch the feature labelled.

Drawing is a skill that only improves with repetition, so practise it often. Our step-by-step guide to drawing biology diagrams is a useful companion for building the habit.

Three different paper styles, a long list of experimental contexts and a command-word vocabulary are easy to underestimate when you are revising alone. It is common to know the content and still lose marks on how you present it. A tutor can mark your practice answers the way an examiner would and show you exactly where marks are slipping away. Building a Past Paper Routine for 0610

Past papers are the most direct rehearsal of the real format, but they work best in stages.

Start topic by topic. Pull questions on one topic, such as enzymes or transport in plants, from several papers and work through them with the mark scheme open to see how marks are awarded.
Move to untimed full papers. Complete a whole paper without the clock, mark it strictly against the mark scheme, and log every lost mark by topic and by type: a content gap, a misread command word, a careless slip or a presentation error.
Finish with timed sets. In the final weeks, sit the multiple-choice and theory papers for your tier under exam conditions, and do the practical paper you are entered for in its own timed slot.

Check that every paper you use is for the 0610 syllabus and your tier. Cambridge also offers a separate IGCSE (9-1) Biology syllabus, 0970, so make sure you are practising the code your school has entered you for. Keep your error log short and specific: "lost 2 marks on a Paper 4 explain question for describing instead of explaining" is something you can fix this week, while "need to revise respiration" is not. If you would like one-to-one help working through the mark scheme, an experienced IGCSE Biology tutor can go through your answers with you and set targeted practice for your tier.

Frequently Asked Questions

How many papers are there in IGCSE Biology 0610?

There are six papers, but each candidate sits three. Core candidates take Paper 1 (multiple choice) and Paper 3 (theory), and Extended candidates take Paper 2 (multiple choice) and Paper 4 (theory). Both tiers then take either Paper 5 (Practical Test) or Paper 6 (Alternative to Practical).

Can you get an A* in IGCSE Biology Core?

No. Core candidates are eligible for grades C to G, and Cambridge states that candidates taught only the Core content can achieve a maximum of grade C. Extended candidates are eligible for grades A* to G, and students aiming for grades A* to C should study the Extended content.

What is the difference between Paper 5 and Paper 6 in IGCSE Biology?

Paper 5 is a Practical Test of 1 hour 15 minutes in which candidates do experiments in a laboratory. Paper 6 is an Alternative to Practical of 1 hour in which candidates are not required to do experiments. Both are worth 40 marks and 20% of the qualification, and both test the same experimental skills.

Can I use a calculator in the IGCSE Biology exam, and what comes after it?

Yes, the syllabus states that calculators may be used in all parts of the exam. After IGCSE Biology, many students continue to Cambridge International AS and A Level Biology, which Cambridge says candidates with grades A* to C are well prepared for. You can read more about A-Level Biology tutoring if you are planning ahead.
